1Cytoprotective effects of Taraxacum species through Nrf2-mediated heme oxygenase-1 expression in murine hippocampal cells

摘要:In order to find new neuroprotective natural products, various Taraxacum species will be collected in the Changbai Mountain. The extracts of these plants will be screened their protective effects in mouse hippocampal HT22 cells. The Taraxacum sp. extract which shows the promising protective effect will be verified its mechanism of action in HT22 cells.Namely, various plant extracts will be tested that the protective effects against glutamate-induced oxidative stress in HT22 cells. Bioactive extract will be verified its cytoprotective mechanism whether it involved the expression of heme oxygenase(HO)-1 or not. HO-1, an inducible stress-response protein, has a pivotal role of protective activity against cellular stress such as oxidative stress to modulate the inflammatory reponse. It is well-known that HO-1 induction is dependent on the expression and activation of Nrf2. Therefore, involvement of Nrf2 in cytoprotective effect of Taraxacum extract will be also evaluated in HT22 cells.

摘要:A high throughput method will develop for rapid screening and identification of bioactive compounds from natural products of Changbai Mountain. The system, integrated with chromatographic separation and UV-MS detection, is compatible with in vitro 96-well microplate or enzyme incubation based bioassays. The stability and applicability of the proposed method will validate by testing radical scavenging capability of a mixture of several known compounds. Moreover, the proposing method will apply to the crude extracts of natural products of Changbai Mountain from which 6 biactive etracts will be screen and characterizing based on their anti-oxidative or anti-obesity or anti-diabetic actibities. Our finding suggest that the integrated system of chromatography coupled with on-line mass spectrometry and microplate based assays can be a versatile and high efficient approach for the discovery of active compounds from natural products.
 
 
3、基于调控HO-1的决明子抗炎活性及机制研究

摘要:血红素加氧酶(HO)-1分解体内血红素生成胆绿素、一氧化碳和铁离子,并与其代谢产物一起发挥着抗炎、抗氧化及细胞保护作用。在体内众多的抗炎机制中HO-1的保护作用日益受到关注。本研究在应用天然药物筛选诱导HO-1的研究基础上,利用天然药物化学和分子药理学的方法,从决明子中提取分离出诱导HO-1的有效部位或活性化合物;并进一步阐明通过诱导HO-1,预防和治疗炎症疾患的作用机制。决明子诱导HO-1的研究,国内外未见报道。
 
4、东北雷公藤中多靶点酪氨酸激酶抑制剂的发现

摘要:东北雷公藤为卫矛科雷公藤属有毒植物,分布于日本、朝鲜以及我国东北,资源较为丰富,研究发现该植物提取物的乙酸乙酯部位有很强的抗肿瘤活性。尤其是对人肠癌细胞和人肝癌细胞,其IC50已分别达到2.65.8μg/mL。基于此,我们拟采用各种现代分离鉴定手段,对产于长白山的东北雷公藤的乙酸乙酯部位进行系统的化学成分研究,同时对分离得到的化学成分进行多靶点酪氨酸激酶抑制活性测试,确定具有多靶点酪氨酸激酶抑制活性的化学成分,从而为新型多靶点酪氨酸激酶抑制抗肿瘤药物的研发提供模式结构和药物前体。
 
5、免蒸红参加工关键技术及其产品有效成分分析

摘要:研究人参双螺杆挤压工艺参数——物料含水量、机筒温度、挤压速度等对挤压制品的红参特定皂苷、美拉德反应产物、红参多糖等有效成分的影响,确定对品质最有影响力的工艺参数,并探讨工艺参数对人参挤压制品有效成分成分的变化机制。通过调控双螺杆挤压工艺参数可以实现人参红参化特有成分的极大化,为免蒸红参加工提供理论依据,人参挤压制品的生理功能研究及其产品深加工提供理论依据。
